No you can't. The description of the Assassination tree makes this clear:

This skill represents your training in the way of assassin. As you progress in this skill, you gain access to deady and precise melee techniques.

Yes. Making long range attacks are nice and could be useful while backstraffing/fleeing. Also with a nice bow or crossbow damage is nice too.

Also, If I might add, some monsters cannot be backstabbed such as slimes. (I did some snooping in the asset pack ;D)

The creatures that doesn't have backs can't be back-stabbed. It would be illogical to stab into something that doesn't exist It would be like making head-shot when your targeted creature doesn't has head.

wow, backstabbing and reach are amazing! missiles are useless as you don't get any bows/xbows until later on.

but backstabbing is so simple in a 2x2 room. just circle them and stab them from behind. It's important to understand that backstabbing is not the classic 'sneak and stab' skill, but simply stabbing a back...

Yeah, backstabbing is one of my favourite parts of Grimrock fighting. And if you have invisibility spell, you CAN actually use it a lot to sneak into backstabbing position for some enemies that are usually hard to do that with due to their speed
